Application
Monoclonal Anti-AFX (FOXO4) antibody can be used for western blot assays at 0.5-1µg/ml. The antibody is also suitable for use in indirect ELISA, immunocytochemistry and microarray applications.
Biochem/physiol Actions
Anti-AFX (FOXO4) antibody, Mouse monoclonal (mouse IgG2b isotype) is derived from the hybridoma AF3.10 produced by the fusion of mouse myeloma cells (NS1 cells) and splenocytes from BALB/c mice. AFX plays a central role in the regulation of cell proliferation. It integrates signals from PI3K/PKB and Ras/Ral signaling pathways to regulate transcription of the cell cycle inhibitor p27kip1. AFX activation results in increased expression of p27kip1, resulting in cell cycle arrest. Overexpression of AFX blocks cells cycle progression at G1 by a p27kip1 dependent mechanism.

General description
Forkhead box O4 (FOXO4) belongs to the FOXO transcription factor family that regulates growth, cell cycle, metabolism, stress tolerance, aging and apoptosis in cells. Studies have reported that USP7 modulates FOXO4 transcription. Moreover, it has been demonstrated that peroxide stress acetylates FOXO4, whereas the longevity protein hSir2SIRT1 deacetylates it.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
