General description

Oxaloacetate (OAA) is an intermediate of both the TCA cycle and gluconeogenesis. It precedes citrate which is formed by the citrate synthase-mediated condensation of acetyl-CoA and OAA. OAA is also formed by the deamidation of aspartate or condensation of CO₂ with pyruvate or PEP. Since mammals do not possess the enzymatic machinery to form TCA cycle intermediates from acetyl-CoA, OAA is one of the anaplerotic entry points via pyruvate and pyruvate carboxykinase. The Oxaloacetate Assay Kit provides a simple, sensitive and rapid means of quantifying OAA in a variety of samples.

Principle

In this kit, oxaloacetate concentration is determined by a coupled enzyme assay, which results in a colorimetric (570 nm)/fluorometric (lex = 535/lem = 587 nm) product, proportional to the oxaloacetate present. Typical detection ranges for this kit are 2–10 nmole (colorimetric) and 0.2–1 nmole (fluorometric).
