Biochem/physiol Actions

Phospholipase C-γ-2 (PLCG2) hydrolyzes phosphatidylinositol-4, 5-bisphosphate to inositol-1, 4, 5 trisphosphate (IP3) and diacylglycerol. IP3 is involved in the increase of intracellular Ca²⁺ levels. Thus, PLCG2 is indirectly involved in increasing Ca²⁺ levels and mediating those pathways which require Ca²⁺. It is also a regulator of many inflammatory pathways. Mutations in the gene expressing PLCG2 have been associated with PLC-γ-2-associated antibody deficiency and immune dysregulation (PLAID). Steroid-sensitive nephrotic syndrome (SSNS) has also been linked to mutations in PLCG2.

General description

Phospholipase C-γ-2 (PLCG2) is a multidomain protein which is expressed in mast cells and B cells. The gene encoding this protein is localized on human chromosome 16.
