Used in the methodology of stable isotope tagging of epitopes (SITE) for identification of virus-induced peptides in vaccine development.

L-leucine-5,5,5-d₃ is used in the quantitative determination of proteins using mass spectrometry and also used in the synthesis of (2S,4R)-(5,5,5-d₃)leucine via stereospecific manner.
