Determine the ability of an organism to split indole from the tryptophan molecule with Thermo Scientific™ Remel™ Spot Indole Reagent. In 1963, Vracko and Sherris used Spot Indole Reagent for the separation of Proteus species and Escherechia coli. In 1969, Lowrance, Reich and Traub found ρ-dimethylaminocinnamaldehyde (DMACA) to be the most sensitive indole reagent which could detect 3 mcg of indole per millilitre of medium.
Spot Indole Reagent is used to determine the ability of microorganisms to split indole from tryptophan molecule by spot test method. Tryptophanases are intracellular enzymes that mediate indole production by hydrolysing the amino acid, tryptophan. This indole combines with DMACA to form a blue-green compound. This is a condensation product formed by an acid splitting of the protein.
