Biochem/physiol Actions
Cereblon (CRBN) is a substrate binding component of the E3 ubiquitin ligase complex. It interacts with thalidomide and its analog to facilitate proteasome-dependent degradation of the transcription factors, Ikaros family zinc finger protein 1 and 3 (IKZF1 and IKZF3). CRBN provides cytoprotection against the ubiquitin-proteasome system-dysfunction. It negatively regulates the AMP-activated protein kinase signaling pathway in vivo. Nonsense mutation of the CRBN gene causes mild type of autosomal recessive nonsyndromic intellectual disability (ID).

Cereblon (CRBN) is encoded by the gene mapped to human chromosome 3p26.2. It is ubiquitously expressed in various subcellular compartments such as nucleus, cytoplasm and endoplasmic reticulum (ER).
